Robin Warren, he observed bacterial cells shaped like curved rods living in the stomach lining of over 100 patients who had ulcers The rod shaped bacteria was identified as Campylobacter pyloridis. Biofilm: It is a layer of attached bacterial, fungal and algal colonies. All the species of microbes are attached with each other and also embedded in an outer surface termed as extra cellular secretion matrix. Which is composed of polysaccharides, lipids, proteins and extracellular DNA.
There are so many method is used for the biofilm detection such as; Tissue Culture Plate (TCP), Tube method (TM), Congo Red Agar method (CRA), bioluminescent assay, piezoelectric sensors, and fluorescent microscopic examination. Bacteria present in biofilm are very infectious to human body and also shows resistance to the antimicrobial drugs.Here we can explain some of the important method:-
Tissue Culture Plate: In this method a diluted broth is prepared using bacterial bio film taken from agar plate containing 10 mL of trypticase soy broth with 1% glucose.Then pour into culture planet and incubated at 37oC. Colony were adhere to the plate surface. Remove the free floating cell by using phosphate buffer. Biofilm of cell set to the surface were fixed by using 2% sodium acetate. Stain the cell with crystal violet(0.1%). Last, take OD at 570nm wavelength.
