Question
In this lab, you will be conducting native agarose gel electrophoresis which is useful in many clinical settings. Both purified proteins and mixed protein samples will be analyzed. One of the samples is purified bovine serum albumin which has an isoelectric point (pl) of 4.7. (2) Would you expect it to have more (+) charged or(- charged amino acids? This protein maintains osmotic pressure of blood while also binding to hormones and ions. Purified Cytochrome C (pl of 10.7) contains a heme group and is involved in electron transport reactions in aerobic cellular respiration. Both albumin and cytochrome C are comprised of single polypeptide chains. (3) Do albumin & cytochrome C have quaternary structure? Egg whites which contains a mixture of proteins, including ovalbumin, and fish muscle samples will also be analyzed. The three dimensional structures of Cytochrome C and Albumin are pictured belowExplanation / Answer
pI = Isoelectric point.
Acidic proteins have low pI.
Basic proteins have high pI.
pI = 4.7 means that the given protein contains greater number of acidic amino acids.
Quaternary structure refers to the interactions among constituent monomeric subunits in a multimeric protein.
BSA is a single subunit protein. Hence it shows no quaternary structure.
Cytochrome C is a single subunit protein. Hence it does not shows quaternary structure.
