Question
Would UV, X-rays, or gamma radiation be a good choice of sterilization method for destroying prions (such as those that cause "mad cow disease")? Explain. 8. 9. You are a staff member in the local hospital burn ward. Why is it extremely important that conditions remairn sterile in this ward compared with other wards such as Maternity or the ER What is the most effective way you can help promote this sterile environment? 10. Before starting mouse surgery, you autoclave your tools at the recommended 121C for 60 minutes. The day after surgery, you notice the mice look sick. They have fever, weakness, listlessness, and some appear to be in pain. However, the tissues you performed surgery on look fine. The same day, a lab co-worker notices bacterial growth in part of the lab. You autoclaved your tools so it cannot be bacteria...can it? What do you think caused your mice to get sick and why? How could you test your hypothesis?Explanation / Answer
8.Sterilising prions requires the denaturation of the protein to a state in which the molecules is no longer able to induce abnormal folding of normal proteins.
Treatment with pro teases ,heat ionising radiation, formaldehyde only decreases the infectivity because prion disease happens due to the misfolding of protein from alpha helical to the beta pledged sheet.
Complete or effective prion decontamination relies upon destruction of the beta structure of the proteins this is done by using chemicals sodium hypochlorite, sodium hydroxide.
9.in burn ward of the hospital conditions remain extremely sterile . Because when burn occurs it exposes the deeper layer of the tissues to the atmosphere inviting micro organisms .. if the first line of defence (skin )is not there then it is much more easy for the micro organism to invade and cause infection and sepsis.
Use of anti microbial chemotherapeutic agents , dressing of wounds with silver gel, aqua cell- Ag.
10.autoclaving has completely sterilised the tools there is no doubt about it ...but there was bacterial growth in part of lab .so this indicates that the spores of bacteria through air has contaminated the wound while performing surgery and resulted in the sickness of the rats.. for conformity just take the samples of the bacteria from rats and from the lab area ... the similarities in the strain and type of bacteria will be one!.
