Answer A) The other type of plasmid that might be present in transformed cells is the self ligated plasmid that do not contain the isolated DNA fragment. Since the site Pst1 is observed on the gene responsible for ampicillin resistance, the cell that grous on both ampicillin plate and tetracyclin plate is having the plasmid that is self ligated while the cell which is growing on tetracyclin but not on ampicillin would be containing recombinant plasmid.
