Syden test is carried for checking dental caris susceptibility. The saliva species are grown with 2 % glucose and bromocresol green (a green dye). When the lactobacillus species are present in the sample when grown in media, the media will change its color to yellow due to decrease in pH due to lactic acid production by bacteria. The dye turns green to yellow at pH below 4.4.
Lactobacillus delbrueckii at 24 hrs. will be slight yellow and at 48 and 72 hrs also it will be yellow.
Saliva sample will give change in color from green to yellow if person is dental caris (tooth decay) due to presence of lactobacillus in saliva which will convert glucose in media to lactic acid droppinng the pH of media below 4.4 at which the bromocresol dye present in the growth media will turn from green to yellow. Hence the color in all tubes will remain yellow due to presence of lactobacillus species in the saliva.
The uninoculated control will remain green only as it doesnt have bacteria to initiate the reaction. Hence dye will remain green only at all three time points.
The above results indicate I have a high susceptibility to dental caris Which is denoted by formation of yellow colour at 24 hr.
