This is the Lineweaver-Burk plot, where, if we extend the lines to the negative x-axis, the points at which they will intersect the axis is the Km for the enzyme. Km is the concentration of substrate at which the enzyme achieves half of the maximum velocity Vmax (the point at which the lines intersect the y-axis is 1/Vmax).
A lower Km, as in case of the dotted line, would imply that very less substrate is required to reach Vmax. This happens when more of the enzyme active site is empty.
A higher Km, as in case of the solid line, implies that the enzyme has low affinity for the substrate, and consequently more substrate is required to reach Vmax. This will happen when the enzyme is getting saturated with substrate and excess substrate is remaining free in the solution.
