Question

Devise/Explain a dilution series for the following:

1) 1:1000, 1:1000, and 1:10,000 dilution of the blended food sample

You have been given a blended 20 g food sample with 180 mL of water. Therefore, the food sample is already a 1:10 dilution.

a) Dispense 0.1 of the blended food sample into a plate for plating. What will the dilution for this plate be? (explain)

b) Dispense 1 mL of the blended food sample into a 99-mL water blank. Using one dilution blank, plate a 1:10000 and a 1:10,000 dilution. Do not make any more dilutions.

Explanation / Answer

1.a) From 1:10 dilution food sample, 0.1 ml is dispensed.

so in 0.1 ml,food sample is=0.1/10=0.01 ml

so the dilution is=0.01:0.1=1:10

b) 1 ml of blended food sample in 99 ml water blank.

so the dilution is=1:100

This dilution blank is added to another 100 ml water to make 1:10000 dilution.
