12- Directly increasing the Na concentration has not much effect since the channel is ligand gated. First a ligand has to bind and that would open up the channel directly increasing the permeability of the channel.

12- Two ways by which neurotransmitter is increased in the cleft :

By inhibhiting the enzymes degrading the neurotransmitter and then increasing its concentration.

Second way is increasing the release of the transmitter from the pre synaptic end either by opening the calcium.channels or by displacing the neurotrasmitters from their vesicles.
