I am attempting to create the Glycolytic pathway (Glc->Pyruvate) in vitro. I purify all the enzymes of the pathway, the co-factors required, NAD+, ADP, ATP and P. I then mix all these components together and add Glucose. I then measure the ratio of Pyruvate:Glucose in the mixture over time. My data is shown in the graph. At the point marked "X" in the graph, I added a 2 substance to the pathway. Time

Here non protein substance added is the GLUCOSE.

Initially you have lots glucose. Once glycolysis process started in vitro the pyruvate get synthesize. This lead to more pyruvate : glucose ratio. you will get exponential increase in the pyruvate : glucose ratio. After some time, glucose in the tube become limited then at that time pyruvate : glucose ratio will be same and have stationary stage.

when we add Substance X in the form of glucose, then once again more pyruvate get synthesize. you will again get exponential increase in the pyruvate : glucose ratio.
