Question

This article to be mentioned is a structure heavy journal article.

Please if you do answer have experience and background in reading published articles and disseminating the info.

These and other articles I will post are critical in my early understanding of genetics. For now I am confused to most of the topic, any simplification would be greatly appreciated.

My question: Can you make it simpler to understand this paper? State the general thesis, and the question the scientists are asking, and how did they approach the problem? Finally the conclusions they reached.

Explanation / Answer

Reverse transcriptase is a multidomain enzyme that is involved in synthesis of DNA from mRNA. therefore its substrate is an oligonucleotide. However, it can be inhibited by number of drugs involving Nevirapine. While the enzyme RT is bound to nevirapine it doesnt bind to its normal substrate oligonucleotide. To elucidate the mechanism of the drug the researchers crystallized the pure enzyme, enzyme with nevirapine and enzyme with oligonucleotide. upon resolution of these crystallized structure, they were able to elucidate the mechanisms of the drug - how it inhibit the normal activity of the enzyme.

the complex structure mentioned in the paper means the compexation of RT with oligonucleotide and drug

in simpler words, without mentioning the domains and sthe subunits, they concluded that the drug binds the enzyme in such a way that it modifies the active site of the oligonucleotide so that the the oligonucleotide cannot bind the enzyme anymore.
