Keeping on ice was necessary because when enzymes are involved in experiments it is better to keep them on ice so that their 3D structure remains intact because three dimensional structure is the key feature of enzyme and keeping it on ice prevents weak forces like hydrogen and intramolecular forces from breaking so that enzyme doesn't loose its function.
