Question

Name two stains that can be used to visualize proteins on a gel after you have run SDS-PAGE. What are their differences, how are they prepped, and what is the de-staining procedure ( if any)?
Name one stain to visualize proteins on a membrane AFTER you have transferred them from gel to membrane and how would you remove the stain to continue on to immunoblotting?

Two gel staining method for protein SDS page.

Coomassive blue staining is easy staining technique, it required only 1 hr for staining and only water as a destaining reagent. Whereas silver staining is tedious takes more time and very sensitive. It required different chemicals for destaining. However silver staining technique is more specific than coomassive blue. We can not use coomassive blue technique if we want to use it for further downstream application (western blot).

Silver staining

General destaining procedure

Ponceau S is sued for membrane staining
