SIM Medium is used to differentiate gram-negative bacteria on the grounds of production of sulfide, indole and motility.

The medium contains sodium thiosulfate and ferrous ammonium sulfate which act as indicators for the generation of hydrogen sulfide. Hydrogen sulfide production is recognized when a black precipitate, ferrous sulfide, is produced when ferrous ammonium sulfate reacts with H2 S gas.

Casein peptone, are other components of SIM Medium which are rich in tryptophan. Organisms which have the enzyme tryptophanase degrade tryptophan to indole. Indole is detected after the addition of Kovacs Reagent which is followed by incubation of the inoculated medium. Indole couples with p-dimethylaminobenzaldehyde and produces a red band at the top of the medium. A negative indole test produces no colour change on the addition of Kovacs Reagent.

agar addition to the medium provides a semi-solid structure which makes the detection of bacterial motility easier. Motile organisms produce turbidity throughout the medium. Non-motile organisms grow only along the stab line and leave the surrounding medium clear.
