Question

Page Gas constant, R = 8.315 J/mol K You are studying an enzyme that follows Michealis-Menton kinetics. The enzyme catalyzes the conversion of S to P (S P) and is inhibited by the competitive inhibitor, Drug. Given the kinetic data in the table below answer questions 1-5 Isl Initial Velocity Drug]- 15 M 3.2 aM/sec 8.9 uM/sec 13.4 HM/see Initial Velocity Drug]-0 11.7 uM/sec 27.5 pM/sec 37.1 HM/sec 45.0 HM/sec 55.4 HM/sec 64.3 uM/sec 72.0 HM/sec 81.8 Usec 87.1 HM/sec 88.7 HM/sec 89.9 1/sec 0.75 2.2 pM 5.0 uM 8.0 HM 12.5M 20 uM 50 1 150 uM 350 M 4.0 mM 25.7 HM/sec 34.6 Lsec 45.0 M/sec 64.3 Usec 79.4 uM/sec 85.1 HM/sec 89.6 M/sec 1. Explanation / Answer

Hi
To solve this its better to do it in excel sheet.
step 1: In excel, type the [S] andinitial velocity drug= 0 and intial velocity drug = 15 in 3 different columns
step 2: Make reciprocal of all these [1/s], 1/d0,1/d15 and type in separte columns
step 3: Plota graph with velocity on Y axis and [S] on X axis. Draw the trendline.
Extrapolate the line on left side, it will touch the Y axis which ic called 1/2Vmax. COntinue extrapolate till it reaches the X axis. This is = -1/km. Reciprocate and you will arrive at Km value.
