1 answer. EcoR1 because EcoR1 is a the restriction enzyme that recognize 5'-GAATTC-3'. When base excision repair corrected the Oxo G then EcoR1 will cut the plasmid and will give two band. But when the repair is not sucessfull EcoR1 will not cut the plasmid.

2 in agarose gel two band of 3.5 kb and 5.5kb will be visible when cut with both EcoR1 and BamH1 and a single band of 9 kb when cut with EcoR1.
