Question

I need to calculate the number of mg/ml that existed in the cuvette for all of these samples. Note that there were two dilution steps for most of these. For samples, 2, 3, 4, and 11, the protein was diluted with water when preparing 1 ml test tube samples. then for all six samples, the protein was again diluted when mixing with Biuret reagent. Calculate the overall dilutions for all 6 samples. Please show the calculation too, I can't figure out how to do this. thanks.

Test tube # 11: .5 ml stock lysozyme (10mg/ml) + .5 ml H2O

Test tube #12: 1.0 ml stock lysozyme (10 mg/ml)

Test tube #13: .5 ml stock DNA + .5 ml H2O

Test tube #14: .5 ml lysozyme (10 mg/ml) + .5 ml stock DNA ( 50 ug/ml)

test tube # 1 2 3 4 5 ml of standard 10mg/ml BSA solution 0 .25 .50 .75 1.00 ml of water 1.00 .75 .50 .25 0

Explanation / Answer

You can use c1v1=c2v2 formula for the calculations

1ml is 1000 microlitres or ul

For test tube 2

10mg/ml*250microlitres = X*1000 microlitres

X= 10mg/ml*250ul/1000ul

= 25/10= 2.5mg/ml

For test tube 3,

10mg/ml*500ul= X*1000ul

X= 5mg/ml

For 4,

10mg/ml*750ul= X*1000ul

=7.5 mg/ml

For test tube 5

X will be 10mg/ml

Test tube 11

10mg/ml*500ul=X*1000ul

= 5mg/ml

Test tube 12 will have 10mg/ml concentration

Test tube 13

The concentration of stock DNA is missing

Test tube 14.

50ug/ml*500ul = X*1000ul

X= 25ug/ml
