1. pUC vector contain MCS(multiclonningsite) in a marker gene LacZ. And insertion of gene into the polylinker will change and interupt the LacZ gene and the enzyme coded (Beta-glactocidase) will not synthesized and the resulting colony will be white confirms that transftransfor is succesful. And anothe marker gene ampr allows colony to grow on amplicilin containing media.
2. Control plates are required to confirmation of appropriate conditions are being provided. If the becteria are unable to grow in control plates then something is wrong with the conditions being provided. Also it provides facility to compare with the supplemented plates.
