Question
1.) You are a comparative physiologist that is interested in understanding somatic growth regulation in a new species of amphibian. Because the new species is very similar to the African Clawed Frog (Xenopus laevis) you assume it must have similar dietary requirements. Given that the nutrient requirements of the African Clawed Frog are known (and readily available), how would you propose to examine the potency of individual essential amino acids in promoting growth (increased protein synthesis/reduced protein degradation) at the cellular level? (20 points) (Note you do not need to list specific lab techniques)Explanation / Answer
We can grow cells extracted from the embryo of the new species in a cell culture. We can grow the cells in medium supplemented with each of the essential aminoacids separately.Allow the cells to grow in incubator or 2-4 days(depends on the growth characteristsics of the cell line).Again depending on the growth of the cells, subculture them.Next cell proliferation assay in each experimental condition which will give an idea of the cell proliferation pattern under each aminoacid. Also, we can isolate protein from each of the cells under different aminoacid conditions exposed. Then run a protein gel to see the protein content and specific proteins that are synthesized in response to each aminoacid supplemented.Increased cell growth would indicate increased protein synthesis and decreased protein degradation. From the gel we can also find out under which aminoacid conditions, more proteins are synthesized. If we have the effect of essential aminoacids on the growth of African clawed frog known , we can compare that with the new species as well.
