Most of the enzymatic reactions are produce or consume hydrogen ions which can cause to change in pH and can denature the enzyme. As we know that the H+ ions is a positively charged which interact with the negatively charged ions. All enzymes are the proteins and the tertiary structures of the proteins are maintained by the hydrogen and ionic bonds between opposite charge of amino acids. H+ ions has ability to interfere with the hydrogen and ionic bonds of the proteins (enzymes) tertiary structure and alter the enzyme structure (by changing the shape of active site). So pH change can leads to change in active site of the enzyme it also affects the enzymatic reaction because substrate shape is complementary to the shape of the enzyme active site. The enzymatic reaction runs smoothly when enzyme substrate complex can form.

All the enzymes works on the specific range of optimum pH, it is important to maintain the pH of the reaction mixture that’s why buffer solution is used.
