Question

1. To generate the growth curve shown below, you transferred cells from an old plate in the refrigerator to a flask of sterile, defined medium that has been optimized for your species. How would the growth curve change relative to the curve shown if you used a medium that was 1 pH unit more acidic than optimal?
a. The slope of the log phase would increase
b. the total cell yield would decrease
c. The lag phase would be shorter
d. There would be no growth at all
e.. The slope of the log phase would decrease

2. To generate the growth curve shown above, you transferred cells from an old plate in the refrigerator to a flask of sterile, defined medium that has been optimized for your species. How would the growth curve change relative to the curve shown if you used an inoculum of exponentially growing cells in the same medium.
a. The slope of the log phase would increase
b. the total cell yield would decrease
c. The lag phase would be shorter
d. There would be no growth at all
e.. The slope of the log phase would decrease

3. To generate the growth curve shown above, you transferred cells from an old plate in the refrigerator to a flask of sterile, defined medium that has been optimized for your species. How would the growth curve change relative to the curve shown if you omit all nitrogen from the medium?
a. The slope of the log phase would increase
b. the total cell yield would decrease
c. The lag phase would be shorter
d. There would be no growth at all
e.. The slope of the log phase would decrease

Log number of viable cells

Explanation / Answer

1. .Ans. Th slope of the log phase will decrease.

Reason: lag phase will be increased because we have used the culture taken out from the refrigerator. Due to which the culture is i stress condition, So it will take longer time in lag phase and hence the log phase will reach very lately, so the slope will decrease.

2. . The slope of the log phase would increase

Due to addition of exponentially growing cells, there would be no lag phase in the culture, and cells will continue to grow exponentially and then leading to death phase.

In this way there would be a longer log phase and a high yield of cell biomass will be obtained.

3.  if you omit all nitrogen from the medium

Then There would be no growth at all because bacteria require carbon and nitrogen as minimalsource of nutrients to grow. Absence of nitrogen will render thye metabolism of the bacteria and hence they will not be able to divide and hence no biomass.
