Question

You have discovered a new area of the brain in the parietal lobe called the Shnoxocampus that seems to be responsible for “selfiness”.
If I wanted to see if taking an excessive number of selfies affected the number of neurons in the area and the dendritic growth of neurons in the Shnoxocampus, what staining techniques should I use? If I then hypothesize that this brain area requires input from the visual cortex, how should I use a tracing technique to confirm the hypothesis? Explanation / Answer

A Nissl body, also known as Nissl substance. iT is a large granular body found in neurons. These granules are of rough endoplasmic reticulum (RER) with free ribosomes. These granules are of a chromophil substance containing a nucleo-protien combined with iron in the cell bodies of neurones. Nissl stain is used to highlight important structural features of neurons. The Nissl substance (rough endoplasmic reticulum) appears dark blue due to the staining of ribosomal RNA, giving the cytoplasm a mottled appearance. Therefore the Nissl staining techniques should be used for the the number of neurons in the Shnoxocampus.

Golgi's method is a silver staining technique that is used to visualize nervous tissue under light microscopy. Dendritic morphology of individual neurons, including dendritic length, arborization and spine number can be assessed by using the Golgi- method of staining.

In neuroscience, anterograde tracing is a research method which is used to trace axonal projections from their source (the cell body or soma) to their point of termination (the synapse). The complementary technique is retrograde tracing. The anterograde and retrograde tracing techniques allow the detailed descriptions of neuronal projections from a single neuron or a defined population of neurons to their various targets throughout the nervous system. These techniques allow the "mapping" of connections between neurons in a particular structure (e.g. the eye) and the target neurons in the brain. Therefore, anterograde tracing technique should be used to confirm the hypothesis that this brain area requires input from the visual cortex.
