Question
2 When genomic DNAs from E. coll (solid line) and Drosophila (dashed line) were individually melted and allowed to reanneal at different Cot values, two different annealing patterns were observed (shown below). 100 : A 50 10 102 10 100 Cot a) Explain why E. coli DNA completely reannealed at 102 Cot, but for Drosophila DNA greater than 100 Cot was needed? 2 pt b) Explain why Drosophila DNA reannealing shows three steps but the E. coli DNA does not? Name the type of DNAs found in each step. 1+3 ptsExplanation / Answer
answer A) this is because ecoli which is a prokaryote has a smaller and simpler circular genomic content compared to drossophila
B) ecoli genome is a single circular DNA while DROSSOPHILA have multiple chromosomes that reanneal at different cots and hence this type of graph is observed. each of the curve shows a chromosome being reannealed.
