Question

Tod do not need to hand in this diagram;just use it to help your understanding of the experiments. 2. The Meselson and Stahl experiment described in class conclusively demonstrated that DNA replication was semiconservative. Predict the results of the Meselson and Stahl experiment for the conservative mechanism of DNA replication. To do so, sketch a plot of IDNAI versus density labeled with representations of the DNA molecules giving rise to the peaks in the plot. In other words, make your answer look like the diagram on p. 52 of the packet. Only show the results after 0, 1, 2, and 3 generations.

Explanation / Answer

Meselson and stahl experiment proved that DNA repliation is a semiconservative process. this was performed by using isotope of nitrogen.

nitrogen isotope N-14 and N-15 (Heavy)

DNA at initial (N-14) --->   DNA replication(N14 & N15) one strand new(N15) -one strand parent strand (N14)------> N14:N15 & N15:N15

This was for semiconservative DNA repliation.

if DNA repliation is conservative means totally new double strand of DNA would form instead of one old and one new strand.

so,

DNA at initial(N14) ----> DNA replication (N14:N14 & N15:N15) -----> 1 ds DNA of N14:N14 and rest will contain N15:N15
