Km is that substrate concentration at which Vmax is half. According to the given plot, Em1 has a Vmax of 0.9 and Em3 has a Vmax of 0.75.

Vmax is half at 0.45; at this value of Vmax the Km comes out to be: Km(Em1) = 10mM

Vmax is half at 0.375; at this value of Vmax the Km comes out to be: Km(Em3) = 10mM

So, you can say about the Km that it is equal for both; Vmax is greater for Em1 than Em3.

According to Michaelis Menton equation:

Km = Vmax [S]/Km+[S]

Note that value of Km and Vmax depends on substrate concentration, and not on the enzyme concentration. So, there will be no effect even if you double the enzyme concentration.

Similarly high Vmax means that the enzyme has high affinity for the substrate.
