7). To investigate the innate immune response to GAS further, macrophages were isolated from mice and cultured with living GAS bacteria. In addition to macrophages from WT mice, macrophages were also isolated from four different strains of knock-out mice: MyD88 --, RIP2--, TAK1--, and ASC-- mice. Macrophages from each strain were cultured with GAS bacteria separately. After 18 hours of co-incubation, the cell- free supernatants from these cultures were tested for their IL-B and TNF- concentrations. The results are show in the figure below. (HINT: MyD88 is required for TLR and IL-1R signaling, RIP2 for NOD receptor signaling, TAK1 for TLR and NOD receptor signaling, and ASC for NLRP3 signaling to activate Caspase-1) IL-1p 4 TNFa 20- 15- 5- type type a). Based on these data, what can you conclude about the function of MyD88 in the innate response to GAS? Explain your reasoning. b). Ans-A- here we can conclude from the above information that the MyD88 is a gene which is required to give information about the production of the proteins which are produced against the foreign invader in our body by the innate immunity system. For interlukins the signals are less developed as compared to wild type while for the toll like factors it is much more than wild type.

Ans-B- in the first graph which is for IL the wild type mRNA is 3.5 ng/ml and MyD88 is 3ng/ml while rest of the mRNA are approx 0.2 ng/ml

In second graph which is for TNF the wild type mRNA is 17ng/ml and for MyD88 is 18ng/ml and for RIP2 and TAK1 it is 2ng/ml. For ASC the mRNA is 18ng/ml.
