Question

16. The figure below shows a series of SNP loci (A-O) loci for two strains of mice (Houston black, Dallas in gray).Which locus would not be useful in mapping novel mutations in a) G b) H c) I d) J e) K MM N N O O Short answer. For each of the questions give a brief answer. If you feel that the answer requires a mathematical expression it is only necessary to put numbers is the proper places, you do not have to calculate the answer 17. You make a cross using an Hfr strain of E. coli that is aziR lys pen and an F strain that is azis lys pen. Interrupted mating studies show that pen enters the recipient last. In an interrupted mating between the same two strains, you select pen recombinants and test by the following genotype distribution: 259 azi lys pen1azi lys pen; 11 azi lys pen*; 29 azis lys What is the gene order (give as first to enter...second...last)?

Explanation / Answer

1) the locus which would not be useful are E, H, J, M, and O. This is due fact that when we do SNP mapping we relly on the RFLP a restriction digestion based method result in fragmentation of DNA.. locus E, H, J, M, and O are giving single band hence cannot predict if it has SNP.

2) gene order is azi- lys-pen. Pen gene is last as it has already been stated in the problem. when scoring 300 individuals, they are finding two genotype which is higher in number same as parental-type however in combination azi(R)-lys(-) -pen(r) has a higher number which suggests azi gene is entering in before the lys gene.
