Question
7. You obtain DNA from three different but closely related species of fish known as species A, B, and C. You take a strand from each species and try to anneal it to the complementary strand from the other species to create hybrid DNA (you monster!). Because the sequences are not exactly complementary, some mispairing occurs. You then try to heat the hybrid samples to see what temperature will cause the hybrid DNA to come apart and obtain the following data: Hybrid Duplex DNA Species A and B Species A and C Species B and C Temperature that the strands separate 80°C 87°C 83oC Based on what you know about hydrogen bonding, which two species have the greatest similarity in their DNA sequence? Why?Explanation / Answer
As we know that the DNA consist of four nucleotide Thymine, Cytosine, Adenine, or Guanine. In which adenine binds to thymine with 2 hydrogen bonds whereas guanine binds to cytosine with making three hydrogen bonds. If we make hybrid duplex of DNA from two species and if there is high complementarity, more hydrogen bonds will form between the nucleotide and more heating temperature will be required to separate those two strands. So, in this question the species A and C are having greatest similarity between them. after that species B and C followed by Species A and B.
