Question
achieve isolation, what might you do differently next time to improve your resuls Not to auser rv Examine the environmental sample. Did you achieve isolation? nok to an 80 er. Were the three different streak methods appropriate to the cell densities recoveredt Most colonies on streak plates grow from isolated colony-forming units (CFUs). On rare occasions, bowever, a colony can be a mixture of two different organisms. If a culture is started from this colony (thinking it is pure), correct identification will be next to impossible because the extra organism could confound the identi- fying test results. How could you verify the purity of a colomy? (The answers may vary depending on what experience you have had prior to performing this exercise.) 1f you found the colony to be a micture of organ isms, what could you do to parify it MICROBIOLOG boratory Theory&Application;, BriefExplanation / Answer
To have pure Colony of one organism, try to Streak the Colony on other plate after checking the contamination on plate. Try to maintain sterility while streaking. To confirm the purity, better would be 16SrRNA sequencing. If culture is mixture of more than one organism then you find peaks within peak. In case of pure Colony, you will find only one peak. By observing the plate carefully you may also able to know the purity of Colony.
