Question
Apply to the following experiment that you have conducted in laboratory. You have grown three following types of E coil cells (one wild-type and two mutants) under each of the following conditions. For the wild-type E. coil cells, the levels of beta-galactosidase (lacZ) produced are as follows: For mutant # 1, the levels of lac Z, produced are as follows For mutant # 2, the levels of !oc Z produced are as follows Based on the experimental results above, which gene was deleted in E. coli cells called knock out mutant #1 and knock out mutant #2? Lac Y (lactose permiase which transports lactose in the cell) Lac repressor CAP cAMP synthetaseExplanation / Answer
In mutant 1 the levels of beta galactosidase is very high in third and fourth condition. In these conditions there is absence of glucose.CAP increase the production of beta galactosidase in the absence of glucose. So the gene deleted in Ecoli is CAP.
In mutant 2 the lac repressor gene is deleted because it halts production of the enzymes encoded by the lac operon. The levels of lacz produced is very less in all four cases.
LacY pumps betagalactosidase including lactose into cell.
