To test whether the mutation in the LEF1 binding site in DNA is sufficient to change LEF1 gene expression, the investigators measured transcription of a “reporter” gene (a gene whose protein product is easy to assay in the laboratory) in cells. Expression of the reporter gene is controlled by 7 repeated copies of the optimal LEF sequence, 7 copies of the LEF1 binding sequence from dark-haired individuals, or 7 copies of the LEF1 binding sequence from blond individuals (see part A of figure below).
