Question

15. (10 points) You become a Crime Scene Investigator and are analyzing one blood sample that has been obtained from an actual crime scene. You are using two SSLP markers (M1 and M2) for genotyping the blood sample. M1 is located in human chromosome 19 and M2 in Y chromosome.

If the results of your genotyping are shown as below, what can you tell police detectives about that blood sample? (a. the blood is derived from how many people --- provide the minimum number; b, how many of these people are males; c, can you rule out the possibility that blood does not include DNA from woman?)

Results:

1. PCR and gel electrophoresis (GE) of M1 SSLP products show five different sizes of DNA fragments (210, 220, 230, 240, 250 bp in length)

2. PCR and GE of M2 SSLP products show two different sizes of DNA fragments (150, 160 bp in length)

Explanation / Answer

These 3/4/5 members could be male or female or both. You cannot rule out the possibility that female is not there.

2. M2 locus will be found only in male. These two different sized DNA fragments show that atleast one male is there. However, two male out of 3/4/5 total members can also be there.
