Question

Using the gel shown below, determine whether the bacterium was transformed, that is, does it contain the plasmid? Briefly explain.

Did recombination occur between the plasmid and chromosome? Briefly explain.

A B C D E F Lane A: EcoRI digest of the bacterial chromosome. Lane B: EcoRI digest of the plasmid. Lane C: HindlII digest of plasmid. Lane D: HindIII digest of the bacterial chromosome. Lane E: EcoRI digest of bacterial DNA after a transformation experiment. Lane F: HindlIl digest of bacterial DNA after a transformation experiment

Explanation / Answer

According to the E and F lanes in gel picture I found both chromosomal and plasmid bands so it is appropriate to say the bacterial cells are transformed.

Yes recombination also possible because upon digestion with EcoRI and HindIII recombinant DNA get digested and forms it's constituent bands.
