Question

Why aorta ring assay is considered as ex vivo experiment? A. Because part of the living tissue (aorta) is isolated from experimental animal for investigation in artificial conditions (e.g. collagen gel) B. Because artificial aorta stent is implanted to the living experimental animal C. Because endothelial cells are isolated from living aorta tissue to culture in artificial conditions D. Because endothelial cells are implanted (injected) to the aorta area of experimental animals Explain ring aorta assay principles Why telomere attrition acts as a tumor suppressor process? A. Because it blocks activity of telomerase B. Because it activates telomerase C. Because it limits the total number of cell division D. Because it blocks apoptosis Describe mechanism of action of telomerase Why PTEN is considered as a tumor suppressor gene? A. It contains protein phosphatase activity B. It prevents cells from intense proliferation by antagonizing P13K activity C. It prevents ce

Explanation / Answer

1) Aorta ring assay is ex vivo experiment because part of the tissue is isoltated from exprerimental animal for investigation in artificial conditions. In this assay, aortic rings are embedded in biomatrix gels and then cultured under chemically defined conditions to generate arborizing vascular outgrowths to study angiogenesis.

2)telomere attrition limits the number of cell division by shortening the telomer length.

Telomerase is the enzyme protein responsible for synthesising telomere. With each cell division, the telomer shortens and when the telomer is completely lost, the cell stops dividing due to lack of telomer which gives the stability to the ends. The enzyme telomerase synthesis telomere and contribute to the continues cell division. This is the main feature of cancer cells.

3) PTEN had phosphatase activity. It is Phosphate and tensin homologue. It acts as a tumour suppresor by stopping cell division and triggering apoptosis. It modifies other proteins by removing phosphate group by its phosphatase activity.
