I answered all the questions already for my bacterial transformation lab. Can someone please help me check if all the answers are correct or not. Really in need of help. I realy am not sure with question 6, please let me know if any answers are incorrect and let me know which is the correct answer. Thank you so much!

Lab 7 Green Fluorescent Protein Questions about Plasmid 1. On the pGLO plasmid, what is the ori sequence for? OA) It is the origin of replication so the bacterial cell can co the plasmid. OB) It allows us to select for bacterial cells that picked up the plasmid. OC) It codes for the green fluorescent protein OD) It allows us to control whether the GFP gene is expressed or not.

For question 5, the answer is A. Arabinose controls the expression of GFP by disabling the repressor. GFP will fluoresce only in the presence of arabinose.

For question 9, plates 1,3 and 4 will show the most bacterial growth, with plate 1 showing highest, as this plate did not select for transformed or non-transformed cells since it has no antibiotic. Thus all the cells, whether transformed or non-transformed, will grow on plate1.

For question 10, the answer is A. There was no GFP because there was no arabinose to disable the repressor. Here, the question of heat or no heat does not arise.
