We talked about an experiment in which proteins were extracted from transformed with the pGLO plasmid and run on a gel. The results are shown in the accompanying figure. The lanes are as follows: Lane 1: Molecular weight markers; Lane 2: No heat; no arabinose; Lane 3: No heat; with arabinose; Lane 4: With heat; no arabinose; Lane 5: With heat; with arabinose. On the gel, why was there no glowing green band in Lane 4? There was no GFP because the bacteria were not exposed to arabinose. The GFP was made but was inactivated because of the heat. There was no GFP because the bacteria were exposed to heat before the proteins were extracted. There was no GFP because the bacteria were exposed to arabinose.

Explanation / Answer

Yes the option 2 is true; the GFP was degraded due to heat. GFP has to incubate at 370C and not more than that.
