Question
You get a job in a new lab with an old centrifuge and your boss asks you to spin tissue culture cells at 1,000xg for 5 minutes. You look at the centrifuge and the control is marked in rpm only. You dig around in the lab and find a ruler and determine that the rotor radius is 270 mm. What speed (rpm) you need the get the required RCF. There is an old piece of lab tape on the centrifuge lid that says don't spin the centrifuge faster that 18,000 rpm. Can you safely spin the cells at l,000g. RCF = 1.12 times 10^-6 * (radius in mm) * (rpm)^2.Explanation / Answer
Answer : To convert rpm into rcf, the following formula is used
g = RCF = 1.12 * 10-6 * radius(mm) * speed2
1000 = 1.12 * 10-6 * 270 * (rpm)2
(rpm)2 = 1000 / 1.12 * 10-6 * 270
rpm = 1818.48
therefore, speed in rpm required to achieve 1000xg is 1818.48 rpm. Since the max speed that could be achieved is 18000 rpm, i.e approx, 97977.6 g , it is safe to spin the cells at 1000g.
