Barley seeds can be divided in two parts; one with the embryo and another part without the embryo. It is alos known that secretion of alpha-amylase initiates seed germination from embryos. Gibberellin hormone induces alpha-amylase initiates seed germination from embryos. Gibberellin hormone induces alpha-amylae gene expression. You know that using potassium iodide, one can detect secreted alpha-amylase. Design an experiment with controls using half barley seeds that will allow to see the role of gibberellin n alpha-amylase secretion. Remember that only embryo containing seed part can germinate so that only part should contain gibberellin hormone.Explanation / Answer
Take the two seed parts i.e. one with the embryo and one without the embryo.
Treat the part with embryo with Gibberellins (GAs) hormone. (experimental group)
Treat the seed part with just the solvent and no Gibberellins (GAs) hormone (this will be the control)
Using the technique of Western blot study the expression of the gene for alpha amylase gene in both the control and the experimental group.
