Question

The map below shows the region around an RJFLP that is associated with Sickle cell anemia. Those with the Mstll site have the wild type beta_A allele, while those with a mutation in the Mstll site (wt: CCTGAGG, mt: CCTGTGG) have the mutant Sickle cell allele beta^s. The arrows at the ends represent primers used to amplify the region (P_L and P_R). This sample is then cut with Mstll and run on a gel to determine the bands present. What would be the predicted size(s) of the DNA band(s) on the gel for someone who is beta^A beta^A ? What would be the predicted size(s) of the DNA band(s) on the gel for someone who is beta^A beta^S ? What would be the predicted size(s) of the DNA band(s) on the gel for someone who is beta^S beta^A ?

Explanation / Answer

Bands on the gel will be 800bp. The DNA sequence will not be cut at all, because there is no site for the restriction enzyme, MstII.
