Question

To study stripe-specific enhancers, researchers have constructed artificial genes in which the enhancer is linked to a reporter gene. In the experiment below a 5-kb fragment is constructed that contains enhancers driving the expression of Eve stripes 2, 3, and 7.

A. Explain how the position of Eve stripe 2 is precisely determined by the products of maternal and gap genes based on the diagram above

B. Which of the above mutants (B) (C) or (D) represents the pattern that would result from a mutation in the binding sites for Giant. Explain your answer

Parasegment 2 Parasegment 3 Parasegment 4 Region of Drosophila embryo stripe numbers (A) NORMAL UTANT (B) M MUTANT (C (D) MUTANT anterior posterior

Explanation / Answer

A) Maternal effect genes like hunchback and bicoid determines the anterior parts of the drosophila embryo through the concentration gradient where as gap genes like hunchback, kruppel and giant are segmentation genes which are regulated by maternal effect genes,so expressed along the anterior - posterior axis like maternal effect genes. hunchback gene acts as a repressor for the expression of giant gene and cannot be expressed without bicoid gene in the anterior - posterior axis. giant activity is required for the formation of anterior border of eve strip 2.
