Question

Describe the experiment with your own words: What is the question the experiment is trying to answer? What is the control? What is the perturbation? Which parameter is measured to reach a conclusion? What is the result? What is the conclusion? How is actin polymerization happening and how is that related to Listeria movement inside the cell? To study the process of actin polymerization, a version of actin with a photo-activatable fluorophore was used. This actin can only be observed by fluorescent microscopy after being activated by light of a certain wavelength. the experiment is summarized in the following diagram. Green represents the actin observed after photo-activation. Q5- Where is the actin polymerization taken place? How do you think actin polymerization can promote Listeria movement/propulsion?

Explanation / Answer

4. The experiment carried out is to prove whether actin polymerization takes place along the bacterial cell surface of Listeria sp.

As a control, an actin inhibitor can be used and the same experiment can be repeated.

The perturbation in the experiment is the use of photo-activable fluorophore.

The movement of the fluorophore is the main parameter to be studied using fluorescent microscopy.

The result obtained is that on a regular time interval of 15 seconds, and 50 seconds, the fluorophore associated with the actin monomer is moving away from the cell surface.

The conclusion is that the Listeria sp induces actin polymerization and this polymerization event initiates on the bacterial surface.

5. The actin monomers are present in mammalian cells cytoplasm. Thus the actin polymerization takes pace in the cytoplasm with the attachment to Listeria sp cell surface.

The actin being motor proteins, the process of actin polymerization aids the Listeria to propel in the host cell and spread the infection.
