Question

Imagine you have three types of cells growing in the lab, one type of cell (Control) has normal hexokinase, and one type of cell (LoF) has a loss-of-function mutation in hexokinase (meaning that it has less or no hexokinase activity), and one type of cell (GoF) has a gain-of-function mutation in hexokinase (meaning that it has higher than normal hexokinase activity). You add the same amount of glucose to the cellular environment of each of the cell types, and measure the glucose in the medium, that is, on the outside of the cells 1 hour later. Assuming you added enough glucose, so that none of the cell types completely used up the glucose, how do you expect the medium glucose concentrations of the cells to compare to each other? Control = GoF GoF LoF

Explanation / Answer

Enzyme hexokinase helps in first step of glycolysis. If its its activity is reduced or increase then glucose metabolism will be affected. As the cell(GoF) has high activity of hexokinase it will consume more glucose than cell(control) and will have less concentration of glucose. Cell (LoF) with reduced hexokinase activity will use less glucose than control cell. Hence concentration of glucose will be more in Cell(LoF). thus medium glucose concentration is expected as

LoF>control>GoF
