Question

1.      For PCR, you want to prepare dilutions of forward and reverse oligos. If you were to prepare each dilution in a new tube such that final concentration of each oligo is 10uM, how much of the each 100uM stock and how much water would you need have in a final volume of 100ul solution to get 10uM primer dilution?

2.If you were to prepare a solution of both primers mixed in together but each still at 10uM in a final volume of 100ul, how much of each stock primer and how much water would you add?

Note: please label and show all steps!

Explanation / Answer

Yoi can use the simple dilution formulae to calculate how much neds to be added:

C1V1 = C2V2

1) For this,

100uM x X = 10uM x 100uL

so, X= 10uL, meaning 10uL of 100uM is needed in 100uL to make 10uM solution so the resy 90uL would be water.

2) since the concentration is not changing,

so forward primer = 10ul

reverse primer = 10ul

and the rest 80uL = water.
