In the first produce the gene is inserted near a gene that is constitutively transcribed. In the second product the gene was inserted near the end of the chromosome adjacent to a telomeric region. A. Which of these two would you expect to be actively transcribed? Why? B. If you were to introduce these genes again, how could you ensure that inserts at both of these locations would be transcribed?

(A) One would expect the gene insert near the constitutively active gene to be transcribed actively as opposed to the insert near the telomerase. Constitutive transcription implies the gene is being transcribed without the promoter ever being switched off. Also, the telomeric region of any chromosome tends to be highly regulated in terms of transcription and barely transcribed. Hence, the gene insert near the constitutvely active pre-existing gene would be actively transcribed.

(B) To ensure that the gene inserted near the telomeric region is also transcribed actively, one can substitute the promoter with a constituve promoter or simply yinsert this gene somewhere upstream altogether with its own promoter.

One could also bring the genes under the influence of an inducible promoter (like lac) and provide induction continuously in order to ensure both genes are produced.
