An experiment was done to determine the maximum growth rate (Hma) and saturation constant ( Ks) of the growth of Escherichia coli. A series of flasks containing fresh and sterile broth are prepared, different initial substrate concentration (So) was introduced to each flask. The same E. coli concentration was introduced with xo 0.3 g/L. The flasks were then incubated for 30 minutes. After incubation the products (P) and substrates (S) concentration were determined, while the cell concentration is measured by using spectroscopy method and stated as absorbance (A). The cell concentration (A) is measured after the cell concentration was diluted with dilution factor of 5.
