Western Blotting is done only after doing the SDS PAGE. The proteins are first separated on SDS PAGE and then the bands are transferred to nitrocellulose membrane to detect the presence of a particular protein of interest with the help of a primary antibody against that protein. The primary antibody is then detected using a secondary antibody which is finally detected through chemoluminescense. That is the whole process of Western Blotting.
