It’s the decoloriser step and if we use too much of the decoloriser agent the gram positive cells become falsely pink and on the other hand if we use too less of it gram negative cells will stay purple and we will get false results.
Mordant is grams iodine in gram staining method and it combines with crystal violet in gram positive bacteria and forms an insoluble complex.
The primary stain is crystal violet which is followed by the mordants step. Next step is decolorisation of iodine with acetone alcohol followed by counter stain with safranin which is a weaker dye than crystal violet . Therefor gram negative remain pink while gram positive purple due to retaining of crystal violet .
