As a clarification on part b: Without any tools or tchniqus rfrs to physical manipulation of said synthtic biology system. It could still be altered theoretically by increasing or decreasing certain variables of the Hill equation. Which variables/parameters should be altered and how in order to create the specified outcome?
3) A constitutive promoter (always on) is used to express a protein from cells grown in a bioreactor a) Derive an expression that describes the steady state concentration of protein [5 points] 150 b) You need to increase protein concentration by 25% but know nothing about synthetic biology and lack the tools to make any genetic intervention. How might you change this system to increase protein concentration by the desired amount? [2 points] Imagine that you now have an engineered strain where the protein half-life has been increased by 50%. How might you fine tune the fermentation process to increase protein concentration by only 25% relative to his initial construct? By how much? [3 points] c)Explanation / Answer
A rather simple approach to increase the concentration would be to just decrease the dilution factors.
For example, if there is excess of a substance, say water, it can be removed quantitively to increase the concentration of the required product.
Given, we need to increase concentration by 25%,
if initial concentration was x, the new required concentration would be 1.25x
Initial dilution was (100-x) (say)
new dilution should be (100-1.25x)
Hence, dilution should be changed accordingly.
