In perfocal microsope the focus the objective remain constant or fixed and while the magnificaiton is changing. For example, if we go for lower to higher magnificaiton like from 10X to 40X or 40X to 10X, for an object, the object will remain in focus and we don't have to adjust the focus each time.

19. there are two types of adjustment knobs in microscope, a coarse and a fine. These two types of knobs are used to adjust the plane of focus so the specimen or object becomes focused and we can see it. Coarse adjust knob is used to move the specimen towards or away from the objective lense. Thus with the help of the coarse adjust knob we can roughly fix the focus plane. Further we can fine tune the focus with the help of fine tune knob.
