Ans) The simple stain process is the one which is used in the labs and are majorly used for finding out the shape of the cell, size and arrangement of structures within the cell. The differential staining technique is the one which allows or enables the observer to distinguish between different types of bacterial cell types based on the color it takes up and is based on the fact that not all cells are able to take up the equal amount of stain.

Ans ) There are couple of advantages of using the differential stain over the simple stain. The differential stain is able to identify the type of micro-organisms or structures of a single organism due to the difference in the stain uptake by different cells. With the basic stain we can only known the physical characteristics of the cell and nothing about the micro-organism. In case of differential staining like gram staining it helps in understanding what type of microbes it is. It helps in identifying chemical and physical properties of the cell.

Ans) Gram staining is the most method for staining the bacteria and is able to identify between gram negative and positive bacteria. The characteristic that helps the bacteria to be stained is the peptidoglycan present in the cell wall. The gram positive and negative bacteria have a peptidoglycan which stains different dye. The gram positive bacteria appear violet in color as the peptidoglycan is able to retain the crystal violet dye. The gram negative bacteria have a very small layer of peptidoglycan which can be dissolved on addition of alcohol and hence it is not able to retain the purple color on washing it with alcohol. On adding a counter stain it retains the pink color which is an identifier of the gram positive bacteria.
