Question

DQuestion 3 1 pts You have a mixture of peptides with the following amino acid sequences TYWSF TEDSE KAGFELCRNEDACER it will take at least two different chromatography columns to separate the three proteins from each other. Which non-denaturing purification methods do you use? Hydrophobic interaction column and a size exclusion coumn O Cation exchange column and a metal affinity column O Size exclusion column and a chitin affinity column SDS PAGE and an anion exchange column D Question 4 2 pts Explain the reasoning for your answer to question 3. Specifically, comment on which techniques will effectively separate which peptides from each other

Explanation / Answer

Answer should be option A. in the first hydrophobic interaction chromatography, TEDSE can be separated from the other two peptides as it does not have any hydrophobic amino acid as compared to others having hydrophobic Amino acids. Although after this also, the two peptides having hydrophobic amino acids will elute out at different time, but peaks may co-incide. Therefore a final size exclusion chromatography will help in separating the two remaining peptides differing greatly in size.
